Core-binding factor beta is required for osteoblast differentiation during fibula fracture healing

Abstract Background Growing evidence has implicated core-binding factor beta (Cbfb) as a contributor to osteoblast differentiation, which plays key role in fracture healing. Herein, we aimed assess whether Cbfb affects differentiation after fibula fracture. Methods Initially, established conditional knockout mouse model for subsequent studies. Immunohistochemical staining was conducted detect the expression of proliferating cell nuclear antigen (PCNA) and collagen II end. Next, isolated cultured osteoblasts from specific mice BrdU analysis, alkaline phosphatase (ALP) staining, von Kossa viability, mineralization, respectively. Western blot analysis reverse transcription-quantitative polymerase chain reaction (RT-qPCR) were used differentiation-related genes. Results The exhibited downregulated PCNA II, reduced ALP activity, well diminished Further, exerted no obvious effects on proliferation. Conclusions Overall, these results substantiated that could promote healing thus provided promising therapeutic target clinical treatment
